need to wait 16 seconds to connect to AJAX-Enabled web sites in Intranet?

First, I don't think Asp.net Ajax will access internet internally.

My opinion is that it's caused by your code that tries to access some resources. Please try a simplest ajax web site in the same environment to see if the problem still exist.
